29D2 Cyl 6 Misfire - change plugs/coil or try again?

Looking for some guidance here...

Running JB4 on Map 5, I hit a 29D2 cylinder 6 misfire on WOT in high gear. Got home, deleted codes, started her back up, all seems OK.

I know that with misfires the typical first step is to replace plug, coil, or both, for that cylinder. My question is - do I have to? Is there any harm in not doing this? Is it possible that this is a one time occurrence, and not the result of a plug or coil that must be replaced?

I changed all plugs 6000 miles ago.

Does it make sense to see if it comes back before acting, or am I just delaying the inevitable?
