There is a difference in sound but it depends on your ear. You can try with a vacuum cleaner or with adhesive tape like body shops do with suction cups on dents.

Try using as-is and see if you hear a difference. Hear it = change it, Don’t hear it = keep it. If you’re the type that can’t stand knowing something isn’t perfect (I fall in this group) then replace it. I think you’ll be fine as-is, though.

It looks like you only got the dust cap.
I believe you can test this before reassembly and then determine. A single speaker isn't that pricey, either.

In all seriousness, I would not worry about that. It may change the sound ever so slightly, but I doubt once the door panel is back on and the engine is running going down the road you'd be able to tell. Besides, you don't know if you did that, it could have been that way since it left the factory.
